A modified orcinol reaction for RNA determination.

چکیده
A MONG the various colorimetric methods for RNA estimation, the orcinol reaction of Mejbaum as modified by Schneider (1) remains the most sensitive and commonly employed technic for purine-bound ribose quantitation. Later attempts that were made to estimate pyrimidine nucleotides and nucleosides (2, 3) have also been based on the color reaction of ribose with orcinol and ferric chloride, which still has the disadvantage of being nonspecific for ribose estimation. The present paper describes a modification which is more sensitive, more rapid, and less affected by DNA, which usually is present in biologic preparations extracted by either trichloracetic acid (TCA) or perchloric acid (PCA). Studies on the Estimation of Deoxyribonucleic Acid and Ribonucleic Acid in Rat Brain
A method for the estimation of DNA and RNA in rat brain is described. The principal modifications of preexisting methods involve initial lipid extraction with chloroformmethanol and continuous stirring during alkaline hydrolysis of RNA and acid extraction of DNA. Comparison of ultraviolet-absorptive methods, phosphate determination, and orcinol reaction for RNA, shows a greater degree of corres...
متن کاملThe fluorometric determination of malic acid.
A sensitive and specific chemical method for the estimation of malic acid in biological materials has long been desired. The condensation of malic acid with orcinol in the presence of concentrated sulfuric acid to form the highly fluorescent homoumbelliferone (7-hydroxyd-methylcoumarin) was reported years ago (1). Although this reaction is relatively specific for malic acid, the quantitative pr...
متن کاملDetermination of nucleic acids in animal tissues.
As reported in a previous paper (l), a new microchemical method for the determination of deoxyribonucleic acid (DNA) has been devised which permits the evaluation of DNA in amounts as small as 2.5 y per ml. The method is particularly useful with very small quantities of tissue. For this reason a study was undertaken to learn the best conditions for the application of the method to the determina...
متن کاملA sensitive method for the determination of ribonucleic acid in tissues and microorganisms.
The increasing interest in the composition of nucleoproteins has emphasized the need for more specific methods of assay of constituent nucleic acids. In a previous paper (1) a method was reported for the assay of deoxyribonucleic acid (DNA) in tissues and microorganisms based on the reaction of p-nitrophenylhydrazine with deoxyribose following hydrolysis of the biological material with 5 per ce...
